While I got about two dozen moments I could list for the Horror Genre (which I will probably post later), in terms of comedy there can only be one candidate...
The hostage moment in particular made me literally puke I was laughing so goddamn hard the first time I watched it
As for drama....fuck it im going all out with the three great final duels of Sergei Leone, with the caveat that you kinda have to really need to watch the movies in order to truly appreciate them (so go do that. Right fucking now. Dollars Trilogy and Once Upon A Time in the West. Will only take you like 12 hours)
Ok. One moment from the OG Terminator movie (my favourite one by a good margin) which helped emphasise how this was easily the darkest movie of the franchise in the best way possible.
To a lesser extent this thematically related scene too
Honestly as much as I enjoy and appreciate Terminator 2 I think the franchise would have done better to stay with the whole "horribly outmatched and outnumbered humans desperately fighting against super powerful machine monsters" thing without having said super powerful machine monsters become cuddly friends of humanity
The hallway fight scene in Inception because I love the fighting in zero gravity look. People can say what they want about the film in general but it took a lot to make this scene look natural and Christopher Nolan pulled it off. I always end up going back just to study each move. I have a feeling in 20 years or so this scene will still hold up because it's done that well.
If I had known about this part I would have watched it on the big screen just to see the hallway fight.

The school hallway scene in Donnie Darko because it was one take and the soundtrack fits so well.
